3M™ Clean-Trace™ UBT 50 Filterable Beverage Kit

Product Description

The 3M™ Filterable Beverage Kit UBT 50 serves to rapidly assess the presence or absence of spoilage organisms in filterable beverages. It can be used with any filterable beverage.

Using 3M Filterable Beverage Kit- UBT 50:

Filter the required volume of beverage (eg beer) through a membrane filter (47 mm). Rinse to remove excess beer residue by filtering 20 ml sterile water through the membrane. Remove the membrane filter and place it on the required selective growth medium. Incubate the plate at the required temperature to enrich organisms and increase sensitivity of the test. Growth of contaminating organisms results in an increase in microbial ATP on membrane surface. After appropriate incubation of the filter (aerobic spoilage organisms 18-24 h, anaerobic spoilage organisms 48-72h) remove Filter aseptically from the media and place it in a sterile Petri-Dish. Pipette 500 µl ATP-ase on membrane surface. Leave it for a minimum of 60 minutes at ambient temperature.

The ATP-ase removes non microbial ATP from remaining product or media from filter surface, whilst microbial ATP remains intact within the cells. Then add 500 µl Extractant Solution to release microbial ATP from within the cells. Swirl the Petri-Dish gently to cover the surface of the membrane and leave Extractant in contact with the filter for about 60 sec +/- 5 sec. Then remove 200 µl of the Extractant and add it to a cuvette containing 100 µl of the Enzyme. Attach the Cuvette Holder to the cuvette and read out immediately in the 3M™ Clean-Trace™ Luminometer. A control test is required to ensure that no contamination of reagents has occured during the assay. It also helps to interpret the results. We recommend a control test with each batch tested.

The kit contains sufficient reagents for 50 tests: • 1 bottle of Extractant - XT • 1 bottle of Enzyme Diluent • 1 bottle of ATP-ase Buffer • 5 vials of B-Reagent. A lyophilized Luciferin/Luciferase reagent in a screw cap vial which, once reconstituted, contains all the components required to generate light in the presence of ATP. • 5 vials of ATP-ase to release ATP from microorganisms in the sample.
